Table 1. EDSTAC's recommended T1 screening assays and possible alternatives

In vitro
  • Estrogen-receptor binding/transcriptional activation
  • Androgen-receptor binding/transcriptional activation
  • Steroidogenesis (to detect substances that interfere with the production of steroids by inhibiting cytochrome P450 enzymes)
  • Placental aromatase (alternative assay to detect substances that interfere with aromatase, an enzyme that converts testosterone to estradiol)
In vivo
  • Rodent 3-day uterotrophic (assay for estrogenicity)
  • Rodent 20-day pubertal female (assay for thyroid and estrogenic effects based on the age vaginal opening occurs in rats)
  • Rodent 5-7-day Hershberger (assay for antiandrogens)
  • Rodent 14-day intact adult male (to detect effects on male reproductive organs that are sensitive to antiandrogens and substances that inhibit testosterone production)
  • Rodent 20-day thyroid/pubertal male (to detect androgens and antiandrogens)
  • Frog metamorphosis (assay for thyroid and antithyroid effects based on tail resorption rate in frogs)
  • Fish gonadal recrudescence (to detect substances that effect maturation in fish)
