Figure 3. PCR-based combinatorial biosynthesis. PCR DNA fragments are amplified from different PKS genes and then ligated together to create new PKS gene clusters. The ligated DNA segments are transferred into bacteria. Next, clones containing the new PKS clusters are isolated as colonies on bacterial plates. Extracts of cultures from bacterial colonies can then be tested for polyketide drug activity.
